Description: The branched-chain a-keto acid dehydrogenase (BCKDH) kinase inactivates the mammalian mitochondrial BCKDH complex by phosphorylation of its decarboxylase component (E1) thus preventing the catabolism of the a-keto acids derived from the essential amino acids, leucine, isoleucine and valine. GloboZymes BCKDH kinase kit is effective for determining the activity of the kinase and to evaluate the effects of test substances on its activity. The kit provides for 50 assays. Assay is based on the reaction of the kinase with BCKDH complex and ATP. The kit includes a directional insert, and contains solutions of MgATP, assay and dilution buffers, and purified sample of BCKDH complex. Store solutions at -70° C. Avoid repeated thawing. [γ-32P]ATP and trichloroacetic acid are not provided with kit.
